The Wallace Rule is a simple, effective heuristic for calculating the melting temperature ($T_m$) of short DNA oligonucleotides in standard salt conditions (typically 1M Na+).

The Formula: $T_m = 2 \times (A + T) + 4 \times (G + C)$. Because G-C base pairs have three hydrogen bonds compared to the two found in A-T pairs, they contribute significantly more to the thermal stability of the duplex.

Application: This tool is widely used in laboratory settings for quick primer design. It provides a reliable estimate to set annealing temperatures for PCR cycles, usually set $3–5^\circ C$ below the calculated $T_m$ to optimize specificity and yield.
