Fluorescence Quenching refers to any process that decreases the fluorescence intensity of a given substance. The **Stern-Volmer equation** quantifies this relationship between the quencher concentration and the loss of fluorescence.

The Equation: $F_0/F = 1 + K_{SV}[Q]$. Here, $F_0$ is the intensity in the absence of a quencher, $F$ is the intensity at a specific quencher concentration $[Q]$, and $K_{SV}$ is the Stern-Volmer quenching constant.

Biophysical Utility: $K_{SV}$ provides a measure of how easily a quencher can access a fluorophore. In protein chemistry, this is frequently used to determine if a residue (typically Tryptophan) is buried in the hydrophobic core of the protein or exposed to the solvent, as only solvent-accessible fluorophores can be effectively quenched by molecules like acrylamide or iodide ions.
