Protein solubility is largely governed by the ratio of polar to non-polar amino acids. A high proportion of hydrophobic residues (e.g., Leucine, Isoleucine, Valine) often leads to the formation of inclusion bodies in bacterial systems, whereas hydrophilic residues promote interaction with the aqueous environment.
Methodology: This tool uses a simplified heuristic: it calculates the frequency of hydrophobic residues relative to the total sequence length. While true solubility depends on tertiary folding and surface charge, this ratio is a reliable first-pass indicator for structural feasibility.
Application: Early prediction saves weeks of laboratory labor. If a protein is predicted to have low solubility, researchers can proactively optimize expression conditions (e.g., lower temperature, chaperone co-expression) or design solubility-enhancing tags (like MBP or SUMO) before beginning production.
