The Isoelectric Point (pI) is the pH value at which a protein carries no net electrical charge. At a pH below its pI, the protein is positively charged; at a pH above its pI, it is negatively charged.

Significance in Purification: Understanding the pI is fundamental for Ion-Exchange Chromatography (IEX). By adjusting the buffer pH relative to the protein's pI, researchers can force the protein to bind to either a cation or anion exchange resin. This technique allows for the selective isolation of a target protein from a complex cellular mixture.

Methodology: This estimation uses the pKa values of the N-terminus, C-terminus, and the side chains of ionizable amino acids (e.g., Histidine, Lysine, Arginine, Aspartic acid, Glutamic acid, Cysteine, and Tyrosine). As pH changes, these residues gain or lose protons, altering the overall electrostatic profile of the protein. Precise pI knowledge is also critical for Isoelectric Focusing (IEF), a high-resolution separation technique used in proteomics to resolve proteins based on their unique pI values.
