The Melting Temperature (Tm) of a DNA primer is defined as the temperature at which 50% of the primer-template duplex dissociates. This is a critical parameter in the Polymerase Chain Reaction (PCR), as the annealing temperature must be carefully optimized relative to the Tm of the primers.

Mechanics of Annealing: The Wallace Rule is utilized here as a reliable estimate for primers between 14 and 20 nucleotides in length. Because G-C pairs are held together by three hydrogen bonds and A-T pairs by two, sequences with higher G-C content possess higher thermal stability, thus requiring higher temperatures to melt.
