Centrifugation is a fundamental process in molecular biology used to separate components of varying densities. The Relative Centrifugal Force (RCF), commonly expressed in 'g', is a measure of the force applied to the sample. Unlike RPM, which is dependent on the specific rotor design, RCF provides a standardized value across different centrifuge models.
The calculation is based on the formula: RCF = 1.118 × 10⁻⁵ × r × (RPM)², where 'r' is the radius in centimeters measured from the center of the rotor to the bottom of the tube. Accurately determining RCF is crucial during cellular fractionation to ensure that organelles like nuclei, mitochondria, or ribosomes are separated without structural damage.
