A hemocytometer is a precision-etched glass slide used to count cells under a microscope. Each large counting square has a known volume (usually 0.1 $\mu L$).

The Calculation: The concentration in cells/mL is calculated as: Density = Average Cells × Dilution Factor × 10,000. The factor of 10,000 accounts for the conversion from $\mu L$ to $mL$ and the volume correction of the chamber.

Experimental Importance: Achieving consistent cell density is critical for experimental reproducibility in mammalian cell culture. Whether preparing for transfection, drug sensitivity assays, or flow cytometry, starting with an accurate cell count prevents variations in nutrient availability, cell-to-cell signaling, and final experimental outcomes.
