A buffer solution resists changes in pH upon the addition of small amounts of acid or base. This is achieved through a mixture of a weak acid and its conjugate base. The Henderson-Hasselbalch equation is the standard model used to calculate the necessary proportions of these components to reach a target pH.
The Equation: By rearranging to $\frac{[Base]}{[Acid]} = 10^{(pH - pKa)}$, researchers can determine the ratio required for a specific buffering environment. Ideally, a buffer is most effective when the pH is within $\pm 1$ unit of its pKa.
Experimental Importance: Biological molecules like proteins and DNA are highly sensitive to pH. Maintaining a precise environmental pH is critical for the stability, solubility, and enzymatic activity of these molecules.
