The purity of nucleic acid samples is evaluated by measuring absorbance at 260 nm (where DNA/RNA absorb) and 280 nm (where aromatic proteins absorb). The resulting A260/A280 ratio serves as an indicator of protein contamination.

Interpretation: * ~1.8: Highly pure DNA. * ~2.0: Highly pure RNA. * <1.6: Significant protein contamination.

Methodology: By dividing the A260 value by the A280 value, researchers can quantify the presence of impurities. Protein contamination can inhibit downstream enzymes like DNA polymerases or restriction endonucleases, making this quality control step a prerequisite for successful molecular biology experiments.
